- Abstract -

Porous Graphitic Carbon is a new stationary phase with good retention properties of very polar analytes due to a combination of two mechanisms: adsorbtion and charge induced interactions of a polar analyte with the polarizable surface of graphite.
We studied the behavior of ondansetron hydrochloride in a chromatographic system using a Hypercarb column and mixtures of acetonitrile and alcohols with acidic modifiers as mobile phase. The research resulted in a new method for quantitative estimation of ondansetron hydrochloride in bulk and dosage forms.

- Abstract -

Topical formulations with corticosteroids are frequently used for the local treatment of inflammatory skin diseases. The aim of this study was to appreciate the correlation between the in vitro release data of hydrocortisone acetate (HA), a mild corticosteroid, from the tested formulations, with the in vivo absorption data and with the anti-inflammatory activity of the tested formulations.
The in vitro release of HA was performed by assessing the HA released from the formulations after a time interval; the skin penetration was evaluated by the degree of depigmentation obtained on the skin after local application of the emulsions. In order to establish the anti-inflammatory activity of the formulations, a clinical study was performed.
The formulation which has the greater percent of HA released has also a better absorption into the dermal layer of the skin, being also the most efficient. The patients treated with this formulation improved their simptomatology (erythema, pruritus) in the shortest time interval, as compared to the other tested formulations.

Il est connu depuis longtemps le fait que la présence dans la molécule de certains groupements chimiques peut etre responsable d'un effet toxique précis. Mais la position dans la molécule de ce groupement, non seulement sa nature, peut moduler cet effet, ce qui a orienté plus récemment l'intéret des chercheurs vers l'étude des relations entre la régio-isomérie et la toxicité.
Les énantiomeres et diastéréoisomeres de molécules chirales bioactives présentent en effet tres souvent une grande différence d'activité soit sur le plan quantitatif (puissance d'action), soit sur le plan qualitatif (différence de nature de l'activité). Nous passerons succinctement en revue certains médicaments et produits chimiques d'intéret : du paracétamol, les dichloro-anilines (DCA), les acides 2-aryl-propioniques, la nicotine etc

- Abstract -

The present study aims to investigate the neuroprotective activity on rat ischemic brain model of several new synthesized hydrazones of N-[1-(2,4-dichlorobenzoyl)-1H-indazol-3-yl-carbonyl]-4-aminobutyric acid with different nitrobenzaldehydes. The brain ischemia was realized by our original technique. The experiment was performed on 30 adult Wistar rats, that received before surgery an unique oral dose of 15 mg/kg from corresponding hydrazone (VI-X). The area of cortical infarctus was observed by means of Zeiss surgical microscope and measured with KS-300 software and by geometrical morphometry. Our previous studies have demonstrated the potential of substituted indazole derivatives as neuronal NO-synthase inhibitors, with promising therapeutic benefit in stroke and neurodegenerative disorders. The efficacy of new synthesized compounds assessed by limiting of cortical infarctus area have shown that only hydrazones derived from 2-nitro- or 4-nitrobenzaldehyde displayed significant neuroprotective properties during acute focal brain ischemia.

- Abstract -

The aim of this paper was that to elaborate a spectrophotometric method of quantitative determination of lisinopril, antihypertensive drug from the class of angiotensin converting enzyme, with a wide usage nowadays.
In this regard, a complexation reaction of lisinopril with copper sulphate was used; the complex combination was analyzed using UV-VIS spectrophotometry. Pure substance of lisinopril dihydrate was used (sponsorship from Medochemie) and 10 mg Ranolip pills (produced by Ranbaxy). The spectrums were recorded in the 500 - 900 nm domain. The calibration curve was traced at 740 nm.
The obtained results prove that the method is sensitive, reproducible and accurate.

- Abstract -

In the perspective of the preparation of some products for topic use standardized in saponins and flavones, for the treatment of peripheric circulatory disorders, cellulite and strech-marks, it is recommended that only the folium- and flores type vegetal products should be harvested from the species Clematis vitalba L.
A high saponin content has been detected in the flower harvested in July (456.99-457.03 g ursolic acid x 10-3 %) a nd in the leaf harvested in September (315.11-315.76 g ursolic acid x 10-3 %). Appreciable flavone content was observed in the folium-type (135.32-135.41 g rutoside x 10-3 %) and flores-type (145.46-145.71 g rutozid x 10-3%) vegetal products harvested in June.

- Abstract -

One method of creating preparations with extended release consists of direct compressions of some mixtures containing active drug substance and various macromolecular compounds, like Hydroxypropyl methylcellulose and Xanthan Gum.
In this paper it was used a Box-Benken experimental design with three variables and three levels (33), which enabled the study of the moisture effect, of the report between polymers Hydroxypropyl methylcellulose / Xanthan Gum and also of the proportion between polymers-active drug substance on diclofenacum "in vitro" release. Dependent variables (responses) of this experimental design were the percentages of active drug substance released at 0.25, 0.5, 1,2,4,6,8,10,12 hours.

The method of obtaining the inclusion complex of enalapril with -cyclodextrin (CD) was based on kneading or kneading followed by coprecipitation. The identity of the obtained supramolecular compound was confirmed by FT IR, differential scanning calorimetry (DSC) and X-ray diffraction. Molecular modeling suggested a supramolecular assembly in which the pyrrolidine group of the enalapril enters inside CD cavity.

- Abstract -

A decrease in reduced glutathione level and of the reduced/total glutathione ratio can be a marker of oxidative stress. A high- performance liquid chromatography with fluorescence detector (HPLC-FLD) method was elaborated for the quantification of reduced and total gluthatione in rat hepatic microsomal preparations, based on derivatization with o-phtalaldehyde.
The method was used to determine the reduced glutathione (GSH) in microsomes treated with cyprofloxacine or norfloxacine, in order to evaluate the potential of fluoroquinolones to induce oxidative stress. A positive control was used consisting in microsomes treated with carbon tetrachloride.
The results of the in vitro experiment showed a statistically significant decrease in the GSH content and of the GSH/total gluthatione ratio, respectively in case of each fluoroquinolones tested and also for the positive control (p < 0.05).

- Abstract -

The quaternary ammonium compounds with thiazolic ring that we synthesized presented in vitro bactericidal properties on Staphylococcus aureus and Escherichia coli stems and antifungic effects on Candida albicans.
Further on, we also studied the effects of these compounds on the Botrytis Cinerea, vegetal fungus which causes the Grey mould disease in plants.
We used the following concentrations for each tested substance: 0.1%, 0.03% and 0.01% in water.
We observed the germination, the growth and the sporulation of the fungi on the culture medium after 3, 6, 9, 12 days. The results were compared with the control medium.
The fungicidal or fungistatic action is in relation with the nature of the substituents on the thiazol ring and, respectively, with the concentrations that we used.

- Abstract -

Diazepam, a benzodiazepine tranquilizer is used for its miorelaxing, anticonvulsivant and tocolitic effects. It has low basic character and the elaboration of new analytical methods for diazepam determination represents a necessity.
This paper proposes a potentiometric method for the determination of diazepam, using an ion selective diazepam electrode. The method was, also, validated. The obtained results show that the chosen method is linear, exact and precise.

- Abstract -

The aim of this experimental work was to study the granulation process of a mixed powder with vitamins and minerals in laboratory scale fluid bed equipment. In order to study the influence of formulation factors on pharmaco-technical properties of granules (Carr Index, Haussner Ratio, time of flow, the granules dimensions, the C.V from the granules dimensions) and pharmaco-technical properties of tablets (C.V. from mass uniformity, chorusing strength, C.V. from chorusing strength, disintegration time, friability) a fractioned factorial experimental design was used. We studied six formulation and process factors: the concentration of the binder, the concentration of the binder solution, atomization pressure, the binder solution spray rate, the shake frequency of the filters bags and the nozzle position.
In order to obtain the best pharmaco-technical proprieties of granules and tablets we calculated the granulation conditions, using Modde 6.0 software (Umetrics, Eweden). The experimental results were close to theoretical ones made using Modde 6.0 software. This means that the theoretical previsions made with design of experiments are correct.

- Abstract -

This study applies the management of marketing principles, in a simple and structured way, to the level of the relation "producer - service supplier". Our analyze points out the "distribution channel" factor, with examples from the Romanian sanitary field. In this regard the study is focused on the relation "producer of drugs - sanitary service supplier".
In the present economical conditions, most producers don't sale the items directly to the final customers; between them there are many intermediaries, named different, with differently functions: merchants (wholesale dealers and retail dealers), agents (errand boys, producer's representations, sales representatives), intermediaries (transporters, banks, advertising agencies).
The paper tries to answer questions like: which is the structure of a distribution channel and what tendency is into this domain?

The paper proposes a method which may be used in practice for evaluation, own-evaluation and co-evaluation. The Performing Management Process has the following stages: the planning of each performance level; the progress, which supposes feed-back and repetition (coaching) the final review (the final evaluation) to analyze the results and plan the next cycle.
The result of the Performing Management (PM) is the link between employee's and company's objectives. Benefits are for both - employee and company - in terms of improvement of productivity (the increase of productivity due to common objectives and performing skills). The evaluation of benefits is made using STAR (Situation-Task-Actions-Results) a constitutive element and tool for the evaluation of the performing management process. The main objective is to analyze the performance regarding a limited period of evaluation (e.g. one year). STAR concept may be presented as training module [1].

- Abstract -

The aim of this work is to separate the enantiomers of propranolol by capillary zone electrophoresis (CZE) in non-aqueous media, using heptakis-(2,3-diacetyl-6-sulfato)-β-cyclodextrin (HDAS-β-cyclodextrin) as the chiral selector. The experiments were performed using a 64.5 cm Ś 50 μm i.d. fused silica capillary column. The applied voltage was 30 kV and the operating temperature of the capillary holder 15oC. The detection was carried out using diode array detection (DAD) by on-column photometric measurements at 230 nm. It was studied the influence of the chiral selector over the resolution. 10 mM concentration of different salts formed by camphorsulphonic acid (CSA), methanesulphonic acid (MSA), formic acid (FA) and chloride anions with Na+, K+ and NH4+ cations were added in the background electrolyte (BGE) as counterions. In order to study the influence of the separation parameters over the resolution of the two enantiomers, a full factorial design was build. The influence of the chiral selector over the resolution was stated. Additives that function as counterions increase the power of separation but it is not obvious how they act and how they influence the resolution.
